PersADE is a comprehensive platform that systematically integrates patient-level context, adverse drug event (ADE) annotations, drug properties and associated human proteins to elucidate the underlying molecular mechanisms. This platform extensively covering the repository of personalized drug-ADE associations (>4 million) and protein-ADE associations, and implements robust confidence frameworks to evaluate molecular mechanisms and reliability of more than 10,000 compounds and 19,000 personalized ADEs.
Basic Search module enables rapid, tri-axial interrogation of the knowledgebase. Users can retrieve curated records by pharmacological entity, by ADE, or by molecular target protein, thereby linking chemical structure, clinical phenotype, and mechanistic biology within a single query.
Patient Search enriches PersADE with individual‐level clinical records, enabling user to filter drug–ADE associations by individual (age, sex, disease, outcomes), pharmaceutic (dosage, formulation, route) information which supports patient-level safety and precision risk assessment.
The Information part provides a structured overview of data provenance and curation workflow for PersADE. By detailing source integration, standardization procedures, and controlled vocabulary mappings, it ensures transparency, traceability, and reproducibility of all records.
PersADE presented a comprehensive repository that included 4,061,772 associations between drugs and personalized ADEs, 31,756 associations between human proteins and personalized ADEs, and 108,677 associations between drugs and human proteins (especially off-targets).
PersADE aggregates 6,980 ADE-associated human proteins across three core granularity tiers: •Function: 5,592 proteins cover all 24 main functions in PENTHER. •Pathway: 348 common KEGG and 6,192 MSigDB pathways. •Drug-Binding: 17,208 validated DTIs with affinity.
•PersADE implements a robust DTA confidence scoring system to evaluate mechanisms for assesses 4,439,661 personalized drug-ADE associations based on 115,000 peer-reviewed references. •PersADE implements personalized confidence and safety score system based on 11,500,749 patients.
PersADE provides multi-parameter search and interactive visualization platform which enables sophisticated queries across ADE nomenclature, chemical structures, molecular targets, and clinical characteristics supported by dynamic network visualization and comprehensive analytical tools.
